Header Image Details

I took this image using a camera that is attached to a microscope.

Non-technical Description:

This is an immunofluorescence image of several yeast cells.  The cells have a protein which is epitope-tagged and I used an antibody to that tag.  This protein is normally found in discrete spots in the Endoplasmic Reticulum (ER) but in these cells, it has been overexpressed and is now found all over in the ER.  The red rings that you see are encircling the nucleus of each cell (because the nuclear membrane is part of the ER).  You can also see ER around the plasma membrane of the cell (called the cortical ER) and tubules connecting the nuclear ER and the cortical ER.

Technical Description:

This is a field of fixed Pichia pastoris cells over-expressing Sec12-Glu-Glu.   The Sec12-Glu-Glu is visualized using an antibody to the Glu-Glu tag followed by a secondary antibody conjugated to Texas-Red.  This image was taken on a Zeiss Axioplan microscope using a Hamamatsu camera and processed using ImageJ and Photoshop software.  The cells are magnified 1000 times.
